Methylxanthines in Chocolate: What You Need to Know
Methylxanthines are natural compounds found in cocoa, coffee and tea. In chocolate, the two that matter most are theobromine and caffeine. They contribute to cocoa’s bitterness and can affect the nervous system, but chocolate does not act like a smaller cup of coffee. The amount and effect depend on the chocolate, the serving and the person eating it.
The three main methylxanthines
The three methylxanthines most commonly discussed in food and drink are:
- Caffeine, most closely associated with coffee and tea
- Theobromine, the main methylxanthine in cocoa
- Theophylline, which is related to the other two but is generally present in much smaller quantities in foods
They share a similar chemical structure, so some of their biological effects overlap. However, they are not interchangeable. Caffeine is the most clearly stimulating of the three in everyday dietary amounts, while the effects of theobromine are more subtle and less certain.
Where do methylxanthines come from?
Methylxanthines form naturally in a limited number of plants, including coffee, tea and cacao. They are part of the plant’s own chemistry rather than ingredients added during chocolate-making.
Once cacao beans are fermented, dried, roasted and processed, some of these compounds remain in the cocoa solids used to make chocolate. Their final concentration can vary with the cacao variety, growing conditions and processing, as well as the recipe used by the chocolate maker.

Does chocolate contain more theobromine or caffeine?
Chocolate usually contains considerably more theobromine than caffeine. One analysis of 32 chocolate couverture and coating samples measured theobromine at about 0.62 to 1.14 milligrams per gram, compared with roughly 0.026 to 0.153 milligrams of caffeine per gram.
That difference is why describing chocolate simply as a caffeinated food misses an important part of its chemistry. Caffeine in chocolate is present at lower levels, while theobromine is normally the dominant methylxanthine.
It is also worth being careful with descriptions of theobromine as a gentler or longer-lasting energy source. In a controlled human study, caffeine reduced tiredness and increased vigour, while theobromine did not consistently improve mood or vigilance. Chocolate may feel uplifting, but taste, texture, sweetness, aroma and expectation all contribute to that experience.
Does dark chocolate contain more methylxanthines?
Generally, yes. Methylxanthines are concentrated in the non-fat cocoa solids, so chocolate containing more cocoa solids will usually contain more theobromine and caffeine.
Dark chocolate therefore tends to have more than milk chocolate. White chocolate is made with cocoa butter but contains no non-fat cocoa solids, so it contains very little by comparison.
Cocoa percentage remains a useful guide rather than a precise measurement. Bean variety, fermentation, roasting and the complete recipe can all affect the final level.
Why do methylxanthines matter to chocolate makers?
Methylxanthines matter because they influence flavour as well as the body. Theobromine and caffeine contribute to cocoa’s natural bitterness and astringency, particularly in darker chocolate.
Bitterness is only one part of the finished taste. It sits alongside acidity, sweetness, aroma, fat and texture. More cocoa does not automatically mean better chocolate: those elements still need to work together so the flavour has depth without becoming harsh.

Can methylxanthines in chocolate affect sleep?
They can, especially for someone who is sensitive to caffeine or eats a larger portion of dark chocolate late in the day.
UK government guidance estimates that a 50g bar of plain dark chocolate contains around 25mg of caffeine, while the same amount of plain milk chocolate contains around 10mg. A mug of coffee typically contains about 100 to 140mg, although amounts vary.
Chocolate contains much less caffeine than coffee, but sensitivity differs. Rather than following one universal cut-off time, notice whether the type, amount and timing of chocolate seem to affect your own sleep.
